Ligation Calculator (Insert to Vector Molar Ratio) by TheCalcTools
How the Ligation Calculator (Insert to Vector Molar Ratio) works
Ligation efficiency depends on the number of molecules, not their mass. Insert mass = vector mass × (insert length ÷ vector length) × molar ratio.
Ratios of 3:1 are typical for standard cloning; small inserts often use 5:1 or more.
The formula
ng insert = ng vector × (bp insert ÷ bp vector) × ratio
Worked example
Using the default values shown in the calculator:
| Vector DNA (ng) | 50 |
|---|---|
| Vector length (bp) | 3000 |
| Insert length (bp) | 1000 |
| Insert : vector molar ratio | 3 |
- 50 × 1,000 ÷ 3,000 × 3 = 50.0 ng of insert.
Result:
| Insert DNA needed (ng) | 50.00 |
|---|---|
| Vector (fmol) | 25.64 |
| Insert (fmol) | 76.92 |
Frequently asked questions
Which ratio should I use?
Start with 3:1 for most inserts. For blunt ends or small inserts try 5:1 to 10:1; check your ligase’s protocol.
